[摘要] 目的:用HPLC-DAD同时测定附子中5种水溶性成分的含量。方法:采用Phenomenex Luna 5u PFP(2)100R(4.6 mm×250 mm,5 mm)色谱柱;流动相为0.1%磷酸-甲醇,梯度洗脱,流速1.0 mL·min-1,柱温35 ℃;检测波长:260 nm(尿嘧啶,尿苷,鸟苷),280 nm(盐酸多巴胺,去甲猪毛菜碱)。结果:尿嘧啶、盐酸多巴胺、去甲猪毛菜碱、尿苷、鸟苷进样量分别在0.004 4~0.44μg(r =0.999 9),0.005 2~0.52μg(r =0.999 9),0.005 2~0.52μg(r =0.999 9),0.010 9~1.09μg(r =0.999 9)和0.007 3~0.73μg(r =0.999 9)范围内有良好的线性关系;平均回收率分别为98.76%(RSD=1.86),102.59%(RSD=1.37%),101.68%(RSD=0.80%),100.06%(RSD=0.51%),102.24%(RSD=0.78%)。结论:本实验首次用HPLC-DAD法对附子中5种水溶性成分进行含量测定,建立的含量测定方法准
